Characterization of JC Polyomavirus Derived from COS-IMRb Cells

JC polyomavirus (JCPyV) causes progressive multifocal leukoencephalopathy (PML), a demyelinating disease of the central nervous system in immunocompromised patients. Study PML-type JCPyV vitro has been limited due to difficulty efficient propagation virus cultured cells. In this study, we carried out long term cultivation COS-7 cells (designated as COS-IMRb cells) transfected with PML type M1-IMRb, an adapted viral DNA rearranged non-coding control region (NCCR). The derived from was characterized by analyzing replication, amount hemagglutination (HA), production protein 1 (VP1), and structure NCCR. HA assays indicated that high present Immunostaining showed VP1-positive were small population represented carrier cell culture. Sequencing analysis NCCR after long-term M1-IMRb conserved without any point mutation. genomic clone COS-IMRb-3 detected Southern blotting single band approximately 5.1 kbp deletion. These findings support may offer useful tool for screening anti-JCPyV drugs.
